What Melanotan II is
Melanotan II is a cyclic, truncated analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), the hormone that signals skin cells to make melanin. It was developed at the University of Arizona, where researchers set out to build a version of alpha-MSH that would survive long enough in the body to study. The ring structure makes it resistant to enzymes that break down the natural hormone. Like the natural hormone, it activates several melanocortin receptors, including MC1R in the skin and MC3R and MC4R in the brain, but it is far more potent and longer-lasting, which is why its research literature spans more than pigmentation.
What the research covers
Melanotan II has a documented clinical-testing history. A 2006 review in the journal Peptides by Hadley and Dorr sets out the record: the linear analogue melanotan I was tested for skin tanning, and melanotan II was tested for pigmentation and for male erectile dysfunction. An effect seen in those trials led to a derivative compound, PT-141 (bremelanotide), which later became an approved medicine. Melanotan I followed its own path to approval as afamelanotide. Melanotan II itself was never approved as a medicine, and its literature also includes published case reports of adverse effects recorded outside clinical trials.
