What is DSIP?
DSIP stands for delta sleep-inducing peptide. It is a chain of nine amino acids, Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, first characterised by Schoenenberger and Monnier in 1977 and named for the slow delta waves of deep sleep it enhanced in their EEG recordings. For a peptide it has an unusual property: a 1994 structural study describes it as a charged, water-loving molecule that can nonetheless diffuse passively across the blood-brain barrier. Our DSIP buying guide covers the vial.
What did the human DSIP sleep studies find?
- Volunteers, 1981. In the first human study, six volunteers (four men, two women) received DSIP or placebo by slow infusion in a double-blind crossover. Median total sleep time rose 59% within 130 minutes, the following night brought shorter sleep onset and better sleep efficiency, and behavioural and EEG testing found no sedation in the classic pharmacological sense. No side effects were observed.
- Chronic insomnia by age, 1986. In 18 people with chronic insomnia, the middle-aged group (29 to 59) reached normal sleep values by the end of a week of DSIP and kept them through a follow-up week. The older group (60 to 83), whose sleep was twice as disturbed at the start, reached normal sleep by the end of follow-up, and the whole sample finished with normal sleep patterns.
- Placebo-controlled, 1987. Fourteen middle-aged people with severe chronic insomnia received DSIP under double-blind, placebo-controlled conditions for seven nights. Sleep improved from the first night and further with repetition, the gain held into the placebo night that followed, sleep efficiency reached the level of normal controls, and daytime alertness and performance rose significantly.
These studies gave DSIP by infusion; the nasal format has not yet been the subject of a published human study.

Frequently asked questions
What does DSIP stand for?
Delta sleep-inducing peptide, named for the slow delta-wave activity of deep sleep recorded in the experiments that first identified it in the 1970s.
How much DSIP is in each spray?
175 mcg. The bottle holds 5 mg of DSIP in 5 mL of sterile saline, about 28 sprays.
What has DSIP research shown?
In a 1981 double-blind crossover study of six volunteers, total sleep rose 59% within 130 minutes of DSIP compared with placebo, and in a 1987 placebo-controlled study of 14 people with chronic insomnia, sleep efficiency reached the level of normal controls.
Which spray stack includes the DSIP nasal spray?
The Sleep & Calm Spray Stack, which pairs the 5 mg DSIP Nasal Spray with the 10 mg Selank Nasal Spray.
Does the DSIP nasal spray need mixing?
No. It arrives ready-mixed in sterile saline in a sealed metered atomiser, so no reconstitution and no bacteriostatic water are needed.
