What is melanotan II?
Natural alpha-MSH breaks down quickly. Melanotan II, also written MT-2 or MT-II, shortens the active core of the hormone to seven amino acids and closes it into a lactam-bridged ring, which makes it far more resistant to the enzymes that clear the natural hormone. At the MC1 receptor it drives the melanin pathway, the basis of its pigmentation research, and it is also active at other melanocortin receptors, which is why it appears in appetite and sexual-response research as well.
A 2006 review in Peptides traces the family: the linear analogue melanotan I was tested clinically for skin tanning, melanotan II for erectile function, and a newer melanotan II analogue, PT-141, was then heading into phase 3 trials. What did the melanotan II studies find?
- Pigmentation, 1996. In a pilot phase 1, single-blind, placebo-controlled study, three male volunteers received melanotan II under the skin on alternating days over two weeks. Two showed increased pigmentation of the face, upper body and buttocks, measured by quantitative reflectance and still visible a week after the study ended. Mild nausea and a stretching and yawning complex were reported.
- Erectile response, 1998. In a double-blind, placebo-controlled crossover study of 10 men with erectile dysfunction of no known physical cause, 8 of 10 developed clinically apparent erections after melanotan II. The mean duration of tip rigidity above 80% was 38.0 minutes against 3.0 minutes on placebo (p = 0.0045), and the transient side effects, nausea, stretching and yawning and reduced appetite, needed no intervention.
Both studies gave melanotan II under the skin; the nasal format has not yet been the subject of a published human study.
