What is KPV?

Alpha-MSH is a melanocortin hormone best known for skin pigmentation, but it also acts on inflammation. KPV is its C-terminal tripeptide, lysine-proline-valine. A 2010 review notes that KPV lacks the sequence needed to bind any known melanocortin receptor, yet retains almost all of the parent hormone’s anti-inflammatory capacity with none of its pigment action. That separation is what makes KPV a research subject in its own right.

What has KPV research found?

• Airway cells, 2012. In immortalised human bronchial epithelial cells, KPV inhibited NF-kB activation, matrix metalloproteinase-9 activity and the secretion of IL-8 and eotaxin, chemokines that recruit immune cells into the airway. The effect went with KPV entering the cell nucleus and blocking the nuclear import of the p65 subunit of NF-kB, and the author concluded that melanocortin peptides offer a robust mechanism for targeting airway inflammation.
• Gut and immune cells, 2008. In human intestinal epithelial cells and human T cells, KPV entered through PepT1, and nanomolar concentrations inhibited the NF-kB and MAP kinase pathways and reduced pro-inflammatory cytokine secretion. Given orally in early-stage laboratory research, it reduced inflammation in two colitis models.
• The research directions, 2010. The same review lists inflammatory skin and bowel conditions, allergic asthma and arthritis among the areas where KPV-type tripeptides are being explored.

The work so far is cell and early-stage laboratory research; KPV has not yet been the subject of a published human trial.
