Description

The Mitochondrial Energy Stack brings together three compounds studied at three different levels of the same organelle — redox environment, membrane structure, and metabolic signalling. The order they are listed in is the order the research logic runs in: each component addresses a condition the next one depends on.

Glutathione is the redox component, and the sequence starts here. A tripeptide of glutamate, cysteine and glycine, it is the cell's principal thiol buffer, and mitochondria maintain their own pool of it — imported from the cytosol, held separately, and responsible for handling the reactive oxygen species that oxidative phosphorylation produces as a by-product. It is also the conjugation substrate that glutathione S-transferases use in phase II metabolism.

SS-31 is the structural component. It binds cardiolipin, the phospholipid that anchors the inner mitochondrial membrane's cristae and electron-transport chain, and is researched for membrane integrity and electron-transport efficiency. It is the most clinically advanced compound in the stack, with human trial programs in rare mitochondrial disease under the name elamipretide.

MOTS-c is the signalling component — a peptide encoded in the mitochondrial genome itself, researched for AMPK activation, metabolic flexibility, and exercise-mimetic signalling. It sits last because it is the adaptive layer, studied on top of the redox state and the membrane structure the first two components address.
