CJC-1295 + Ipamorelin: Research Reference

CJC-1295 + Ipamorelin is a blend of two synthetic growth hormone secretagogues in one lyophilised vial: CJC-1295 without DAC, a 29-residue modified fragment of growth hormone releasing factor also called modified GRF (1-29), and ipamorelin, a five-residue peptide that acts at the ghrelin receptor. The Vistara Labs 10 mg vial holds 5 mg of each.

This reference collects what a laboratory needs to know about the blend as a material: how each sequence behaves in the vial, what they dissolve in, how the vial is handled at the bench and what to do when an order arrives. Each section below has its own page.

CJC-1295 (no DAC) data are from PubChem CIDs 91976842 and 56841945; ipamorelin data from PubChem CID 9831659 and the Raun et al. sequence below.

Three features of these sequences explain most of the blend’s behaviour. CJC-1295 differs from native growth hormone releasing factor (1-29), listed in PubChem as sermorelin (CID 16132413), at four positions: D-alanine at 2, glutamine in place of asparagine at 8, alanine in place of glycine at 15 and leucine in place of methionine at 27. Two of those swaps remove the native fragment’s most reactive residues, Asn8 and Met27. Ipamorelin is built from protected parts: aminoisobutyric acid at the front, two D-amino acids in the middle and an amidated end (Raun et al., European Journal of Endocrinology, 1998). And CJC-1295 is the longer, more hydrophobic and more reactive of the two, so it sets the pace for the blend in solution.

What it is studied for

The two peptides reach the same pituitary cells through different receptors. CJC-1295 was developed as an analogue of human growth hormone releasing factor (1-29) that acts at the GRF receptor; the original paper describes it as tetrasubstituted, with an added albumin-binding lysine that defines the DAC form (Jetté et al., Endocrinology, 2005). In healthy adults the DAC form gave an estimated half-life of 5.8 to 8.1 days (Teichman et al., Journal of Clinical Endocrinology and Metabolism, 2006). The no-DAC form in this blend lacks that albumin-binding group, and most published human work used the DAC form.

Ipamorelin was described as the first selective growth hormone secretagogue: it released growth hormone in rat pituitary cells, anaesthetised rats and swine without raising ACTH or cortisol at the levels GHRP-6 and GHRP-2 did (Raun et al., European Journal of Endocrinology, 1998). A modelling study in healthy volunteers measured a terminal half-life of about 2 hours and a single episode of growth hormone release (Gobburu et al., Pharmaceutical Research, 1999). Our comparison with a related compound is in CJC-1295 + Ipamorelin vs tesamorelin.

Stability

Ipamorelin has very few weak points. CJC-1295 has three glutamines, two aspartates and two tyrosines, plus a hydrophobic helical stretch, so it is the component that limits the blend. The CJC-1295 + Ipamorelin stability page walks through both sequences, freeze-thaw, light and what visible changes mean.

Solubility

Both peptides carry a net positive charge in bacteriostatic water, so the blend dissolves in it without acid, though CJC-1295 can take a little longer to clear. The CJC-1295 + Ipamorelin solubility guide explains why and what causes haze.

Handling

Bench practice for the blend is about patience while it dissolves, sterile access and cold, labelled storage. The CJC-1295 + Ipamorelin handling page lists the steps and the common mistakes.

On arrival

What a sealed vial should look like, why it travels without an ice pack and where it goes first are on the CJC-1295 + Ipamorelin on-arrival page.

Storage and reconstitution

Fridge and freezer windows, before and after mixing, are on how long CJC-1295 + Ipamorelin lasts. Water volumes and the concentration table for the 10 mg vial are on how much bacteriostatic water for a CJC-1295 + Ipamorelin vial.

FAQ

Short answers to the questions researchers ask most about storing, mixing and handling the blend are on the CJC-1295 + Ipamorelin FAQ.

The no-DAC single vial

CJC-1295 (No DAC) 10 mg is the same modified GRF (1-29) supplied on its own, so the GRF-side component can be set independently of the ghrelin-side one. Everything this reference says about the CJC-1295 half of the blend applies to it: it is the less stable of the two peptides, so the single vial follows the same cold, dark, sterile handling and the same storage windows.
