Tesamorelin: One Small Change to a Natural Hormone

Tesamorelin is a modified version of a hormone your body already makes. The natural molecule, growth hormone releasing hormone, is broken down in circulation within minutes. The problem tesamorelin was built to solve
Growth hormone releasing hormone (GHRH) signals the pituitary to release growth hormone. As a research tool it has an awkward property: an enzyme called DPP-4 clips it almost immediately, which makes the native peptide difficult to study over any useful interval.
Tesamorelin is GHRH(1–44) carrying an added trans-3-hexenoyl group at one end. That addition obstructs the enzymatic cleavage site, so the molecule persists longer than the unmodified hormone while acting at the same receptor.
This is a recurring pattern in peptide research: take an endogenous signalling molecule, make a small structural modification that resists degradation, and you get something you can actually run an experiment with.
How it differs from the secretagogues
Tesamorelin and compounds like ipamorelin both end up influencing growth hormone signalling, but they arrive by different routes. Tesamorelin is a GHRH analogue — it acts at the GHRH receptor. Secretagogues such as ipamorelin act at the ghrelin receptor instead.

Form and handling
Tesamorelin is supplied as a lyophilised powder in a sealed vial. Like most peptides in this class it is reconstituted in a laboratory setting with bacteriostatic water and kept refrigerated and shielded from light thereafter. The dry vial is the stable form; once in solution, the clock starts.
