What is Epitalon?

Epitalon is the synthetic tetrapeptide Ala-Glu-Asp-Gly, often shortened to AEDG. A St Petersburg research group designed it from the amino-acid composition of epithalamin, a peptide extract of the pineal gland they had studied since the 1970s, and the first Epitalon papers appeared in 2000.

Two names cause most of the confusion. Epitalon and Epithalon are two spellings of the same synthetic peptide, so a search for Epithalon in Canada leads to the same vial. Epithalamin is the older pineal extract it was modelled on, and when a paper reports long human follow-up, it is usually describing epithalamin.

What has Epitalon research shown?

The strongest Epitalon findings come from human cell cultures, with a smaller set of human reports alongside.

• Telomerase switched on, 2003. In human fetal fibroblasts that normally lack telomerase, adding Epitalon induced the enzyme’s catalytic subunit, raised telomerase activity and lengthened telomeres, the protective caps on the ends of chromosomes.
• Past the division limit, 2004. Fetal lung fibroblasts that had stopped dividing at passage 34 regained telomere length comparable to early passages with Epitalon, made 10 extra divisions to passage 44 and were still dividing.
• Independent replication, 2025. A Brunel University London group found that Epitalon lengthened telomeres in normal human epithelial cells and fibroblasts by raising hTERT expression and telomerase activity. In two breast cancer cell lines the lengthening ran through a separate, telomerase-independent route known as ALT.
• Melatonin and the body clock, 2020. In middle-aged people with reduced melatonin output, AEDG raised urinary excretion of the main melatonin breakdown product 1.7-fold and normalised over-expression of the clock genes Clock and Csnk1e 1.9 to 2.1-fold.
• The retina, 2002. In patients with degenerative retinal disease (retinitis pigmentosa), the authors reported a positive clinical effect in 90% of cases.

The parent extract has the longest human follow-up: in a 2003 report of 266 older adults followed for 6 to 8 years, mortality was 1.6 to 1.8-fold lower in the epithalamin group than in the control group.
