Kisspeptin Side Effects: What the Research Reports

Sexual Health & Intimacy Research

Across the published human studies, kisspeptin has been well tolerated with no adverse effects reported: in two randomised, placebo-controlled trials that each followed 32 adults with low sexual desire, a 75-minute intravenous infusion produced no side effects, and in men it had no significant effect on blood pressure or heart rate. In IVF research, a kisspeptin trigger in 60 women at high risk of ovarian hyperstimulation produced no moderate or severe cases. This guide sets out what each study recorded and how Kisspeptin and the ready-mixed Kisspeptin Nasal Spray fit a laboratory research setting.

Findings

  • In 32 women with hypoactive sexual desire disorder, kisspeptin infusion was well tolerated with no reported adverse effects (Thurston et al., 2022).
  • In 32 men with the same condition, there were no side effects or adverse events and no significant clinical effects on blood pressure or heart rate (Mills et al., 2023).
  • Among 60 IVF patients at high risk of ovarian hyperstimulation syndrome (OHSS), egg maturation occurred in 95% and no woman developed moderate, severe or critical OHSS (Abbara et al., 2015).
  • Women with hypothalamic amenorrhoea who received kisspeptin twice weekly for 8 weeks had raised reproductive hormones and no adverse effects (Jayasena et al., 2010).

What is kisspeptin?

Kisspeptin is a neuropeptide made in the hypothalamus that is a key regulator of the reproductive hormone system. It stimulates the release of gonadotrophin-releasing hormone (GnRH), which in turn drives the pituitary to release luteinising hormone (LH) and follicle-stimulating hormone (FSH). It circulates in several lengths, and human studies have used kisspeptin-54 and the shorter kisspeptin-10. In healthy men, kisspeptin-10 produced a rapid rise in LH, and longer infusions raised testosterone and the frequency of LH pulses (George et al., J Clin Endocrinol Metab, 2011). The background is in What Is Kisspeptin? and the work in men in Kisspeptin and Testosterone.

What did the human studies report?

Much of the human research comes from one London group and used intravenous infusion or injection under the skin, under clinical supervision. Safety was recorded in each study, and the picture is consistent.

Sexual desire trials, 2022 and 2023

In a double-masked, placebo-controlled crossover trial, 40 premenopausal women with hypoactive sexual desire disorder (HSDD, persistently low desire that causes distress) were randomised and 32 completed both visits. Kisspeptin modulated activity in brain regions involved in sexual and attraction processing, and it was well tolerated with no reported adverse effects (Thurston et al., JAMA Netw Open, 2022). A matching trial in men with HSDD randomised 37 and 32 completed. Kisspeptin significantly changed activity in the brain’s sexual-processing network and increased penile tumescence in response to sexual stimuli by up to 56% more than placebo. The authors report no side effects or adverse events, and no significant clinical effects on blood pressure or heart rate (Mills et al., JAMA Netw Open, 2023). Both trials are discussed in Kisspeptin for Libido.

IVF trials

Ovarian hyperstimulation syndrome is a potentially serious complication of IVF, linked mainly to the hormone used to trigger final egg maturation. In a phase 2 randomised trial of 60 women at high risk of OHSS, a single kisspeptin-54 injection triggered egg maturation in 95% of women, live-birth rates per transfer reached 45% across all dose groups, and no woman developed moderate, severe or critical OHSS (Abbara et al., J Clin Endocrinol Metab, 2015). A later cohort comparison of high-risk women found OHSS symptoms were least frequent after kisspeptin; compared with kisspeptin, the odds of an OHSS diagnosis were 33.6 times higher after hCG and 3.6 times higher after a GnRH agonist trigger (Abbara et al., Clin Endocrinol, 2018). The two hormones are compared in Kisspeptin vs HCG.

Blood pressure and heart rate

Researchers also checked cardiovascular effects. In healthy men and women monitored for four hours after kisspeptin-54, there were no significant changes in heart rate or blood pressure (Nijher et al., Br J Clin Pharmacol, 2010).

What does research show about longer-term use?

The longest course in these studies ran eight weeks. In women with hypothalamic amenorrhoea (absent periods caused by reduced hypothalamic signalling), twice-weekly kisspeptin-54 significantly raised reproductive hormone levels compared with saline, and no adverse effects were observed. The same study mapped how often the hormone system can be stimulated: with twice-daily injections, responsiveness faded, with the FSH response nearly gone by day two and the LH response declining more gradually, whereas twice-weekly injections produced only partial desensitisation (Jayasena et al., Clin Pharmacol Ther, 2010). This loss of response with frequent exposure, called tachyphylaxis, is a property of the reproductive axis rather than a side effect, and it informs how kisspeptin studies are designed. How long kisspeptin circulates is covered in Kisspeptin Half-Life.

How does kisspeptin compare with related compounds?

Kisspeptin works upstream, on the brain’s reproductive hormone signal, whereas PT-141 acts on melanocortin receptors and has a well-characterised side-effect profile led by nausea and flushing; see PT-141 Side Effects and PT-141 vs Kisspeptin.

Why research-grade quality and storage matter

The trials above used characterised, clinical-grade peptide, so verified identity and purity are what let laboratory results be compared with that work. Vistara Labs supplies Kisspeptin as a lyophilised vial and as a ready-mixed metered nasal spray. Every product is third-party tested, with certificates on the lab reports page. Mixing is covered in the kisspeptin reconstitution guide and stability in How Long Do Peptides Last After Mixing?

Kisspeptin is supplied by Vistara Labs for laboratory research use only. The human studies above were clinical research carried out under medical supervision.

Frequently asked questions

What side effects has kisspeptin research reported?

The published human studies report kisspeptin as well tolerated. Trials in 32 women and 32 men with low sexual desire recorded no adverse effects, and an eight-week study in women with hypothalamic amenorrhoea observed none.

Does kisspeptin affect blood pressure?

In healthy men and women monitored for four hours after kisspeptin-54, there were no significant changes in blood pressure or heart rate. The 2023 trial in men also found no significant clinical effects on either.

What did kisspeptin IVF research show about safety?

In 60 women at high risk of ovarian hyperstimulation syndrome, a kisspeptin-54 trigger matured eggs in 95% and no woman developed moderate, severe or critical OHSS. A later comparison found OHSS symptoms least frequent after kisspeptin.

What did longer kisspeptin studies show?

Twice-weekly kisspeptin-54 for eight weeks raised reproductive hormones in women with hypothalamic amenorrhoea with no adverse effects. Twice-daily injections led to a fading hormone response, known as tachyphylaxis.
