What is kisspeptin?

Kisspeptin is a naturally occurring peptide hormone encoded by the KISS1 gene. The gene was first described in 1996 by researchers at Penn State in Hershey, Pennsylvania, who found it expressed only in melanoma cells that did not spread. In 2001, peptides of 54, 14 and 13 amino acids sharing a common RF-amide end were isolated from human placenta, shown to activate the receptor GPR54 and named kisspeptins. Kisspeptin-10 is the minimal kisspeptin sequence with full intrinsic bioactivity, while kisspeptin-54 is the major circulating form in humans.

The reproductive role emerged in 2003, when mutations in GPR54 were found in people who had never gone through puberty (idiopathic hypogonadotropic hypogonadism). What has kisspeptin research shown?

• Men, 2005. In a double-blind crossover study of 6 men, a 90-minute intravenous infusion of kisspeptin-54 significantly increased LH, FSH and testosterone compared with saline.
• Women, 2007. Subcutaneous kisspeptin-54 raised LH in every phase of the menstrual cycle, with the greatest effect in the preovulatory phase: a mean rise of 20.64 IU/L over baseline.
• Kisspeptin-10, 2011. In healthy men, a kisspeptin-10 infusion raised mean LH from 5.2 to 14.1 IU/L and LH pulse frequency from 0.7 to 1.0 pulses per hour.
• IVF, 2014. In 53 women undergoing IVF, a single subcutaneous injection of kisspeptin-54 induced egg maturation; clinical pregnancy followed in 23% (12 of 53).
• IVF at high risk of OHSS, 2015. In 60 women at high risk of ovarian hyperstimulation syndrome, egg maturation occurred in 95%, live birth per transfer was 45%, and no woman developed moderate, severe or critical OHSS.
• Sexual and bonding images, 2017. In 29 healthy young men, kisspeptin enhanced limbic brain activity in response to sexual and couple-bonding images and attenuated negative mood.
• Women with low sexual desire, 2022. In a placebo-controlled trial of 32 premenopausal women with hypoactive sexual desire disorder, kisspeptin modulated sexual and facial attraction brain processing, with no reported adverse effects.
• Men with low sexual desire, 2023. In a placebo-controlled trial of 32 men with hypoactive sexual desire disorder, kisspeptin significantly modulated sexual brain processing and increased penile tumescence to sexual stimuli by up to 56% more than placebo.
