- 10mg PT-141 Nasal Spray, 2004: in two double-blind, placebo-controlled studies of men, intranasal PT-141 peaked in the blood at a median of 30 minutes, and at the higher amounts tested the erectile response was statistically significant against placebo (International Journal of Impotence Research).

Why pair PT-141 with oxytocin?
The two peptides answer different questions in libido research, and both were studied mostly as nasal sprays.
- Desire and arousal. PT-141, known clinically as bremelanotide, is an alpha-MSH analogue that binds the MC4 receptor with high affinity and also acts at MC3, receptors that sit mainly in the central nervous system. Its research therefore centres on brain pathways rather than circulation. In 2006, 18 premenopausal women with female sexual arousal disorder each received one nasal administration of PT-141 and one of placebo, and more reported moderate or high desire after PT-141.
- Trust and attachment. Oxytocin is released into the blood from the posterior pituitary and acts on a single receptor. Its intranasal record goes back decades: a commercial nasal spray was in use by 1960, and in 2005 a single nasal dose raised trust between people in a Nature study.
