What is Semax?
Semax is a synthetic heptapeptide, Met-Glu-His-Phe-Pro-Gly-Pro. Its first four residues come from the ACTH 4-10 region of adrenocorticotropic hormone, and the Pro-Gly-Pro tail was added to slow enzymatic breakdown, the usual weakness of short natural fragments. None of the adrenal-stimulating activity of full-length ACTH survives; the interest lies in neurotrophic signalling. It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and has decades of clinical use in Russia as nasal drops. Most published work comes from Russian groups and centres on stroke and brain-derived neurotrophic factor.
What is Selank?
Selank, Thr-Lys-Pro-Arg-Pro-Gly-Pro, begins with tuftsin, a naturally occurring immune-modulating fragment of immunoglobulin G, and ends with the same stabilising Pro-Gly-Pro extension used in Semax. It was developed at the same Russian institute. Beyond the 2015 phenazepam study, a 2008 study recorded shifts in immune-system cytokine markers in patients with generalised anxiety, and laboratory work examines GABA-A receptor signalling and the enzymes that break down enkephalins.
What is Cerebrolysin?
Cerebrolysin is made by controlled enzymatic breakdown of purified pig brain proteins, which leaves a defined mixture of low-molecular-weight neuropeptides and free amino acids. Its activity is attributed to the mixture as a whole, with research centred on neurotrophic, growth-factor-like signalling. Alongside CARS, the CAPTAIN I trial in moderate-to-severe traumatic brain injury (2020) reported significantly better results on several thinking and memory measures in the Cerebrolysin group. Results vary by condition and outcome measure. The vial sold here is a lyophilised laboratory material, separate from the clinical solution used in European and Asian stroke units.
Why are Semax, Selank and Cerebrolysin studied together?
The three compounds cover different arms of the same neural machinery. Semax is the activating arm, studied around BDNF and plasticity. Selank is the inhibitory arm, studied on GABAergic tone and the stress response. Cerebrolysin supplies the broad neurotrophic base.
