What is PT-141?
PT-141 is a cyclic seven-amino-acid peptide built on the core of alpha-MSH, the melanocortin hormone. It is an agonist at the MC3 and MC4 melanocortin receptors, and its research centres on the central pathways of sexual desire and arousal. It grew directly out of melanotan II research, a lineage traced in Melanotan II and the PT-141 connection.
What did the nasal PT-141 studies find?
- Men, 2004. In two double-blind, placebo-controlled studies of intranasal PT-141, in male volunteers and in men with mild-to-moderate erectile dysfunction, blood levels peaked at a median of 30 minutes and the half-life ran 1.85 to 2.09 hours. At the higher amounts tested, the erectile response was statistically significant against placebo, with the first erection at about 30 minutes. It was well tolerated, and no maximum tolerated level was reached.
- Women, 2006. Eighteen premenopausal women with female sexual arousal disorder each received a single nasal administration of PT-141 and a placebo, in random order. More reported moderate or high sexual desire after PT-141 (P = 0.0114), and among women who attempted intercourse within 24 hours, significantly more were satisfied with their arousal (P = 0.0256).
- Tolerability, 2017. In a phase 1 crossover study of 24 adults, 12 men and 12 women, single nasal administrations taken with or without alcohol were safe and generally well tolerated, with no clinically significant pharmacokinetic interaction and no significant drop in blood pressure.
The later phase 3 programme moved to a form given under the skin: two 2019 trials randomised 1,267 premenopausal women with low sexual desire and found significant gains in desire against placebo (P < .001). The full trial history is in PT-141 human studies.
