Selank vs SSRIs: How They Differ
Selank is a seven-amino-acid peptide built on tuftsin and studied in anxiety disorders, while SSRIs (selective serotonin reuptake inhibitors) are a class of approved antidepressant medicines that block the serotonin transporter and are used for depression and anxiety disorders. The core difference is the target: SSRIs act on serotonin reuptake, while Selank’s research points to enkephalin-degrading enzymes and allosteric modulation of GABA binding. Their evidence also differs in scale, from hundreds of SSRI trials to a small set of Selank clinical studies. Here is how they compare.
Findings
- SSRIs inhibit presynaptic reuptake of serotonin at the serotonin transporter, raising serotonin at the synapse; six structurally unrelated SSRIs share that mechanism (2021).
- A network meta-analysis of 522 trials and 116,477 adults with major depression found every one of 21 antidepressants more effective than placebo (2018).
- For generalised anxiety disorder, a network meta-analysis of 89 trials and 25,441 patients found escitalopram, sertraline and fluoxetine efficacious against placebo (2019).
- Selank’s anti-anxiety effect matched the benzodiazepine medazepam’s in 62 patients with generalised anxiety disorder or neurasthenia, with added anti-fatigue and stimulant effects (2008).
- In a 2014 study of 60 patients, Selank’s anti-anxiety effect lasted a week after the last administration.
What are SSRIs?
SSRIs became a mainstay of psychiatric treatment after fluoxetine was introduced in the United States in 1988. Their primary mechanism is to inhibit presynaptic reuptake of serotonin at the serotonin transporter, which increases serotonin at the postsynaptic membrane. The six major SSRIs marketed in the United States are fluoxetine, citalopram, escitalopram, paroxetine, sertraline (Zoloft) and fluvoxamine: structurally unrelated molecules with a shared mechanism, but each with its own pharmacokinetics and side-effect profile (Edinoff et al., Neurol Int, 2021).
SSRIs act on the transporter directly, yet the clinical effect of most antidepressants arrives with a delay. That gap is one reason researchers look beyond a purely neurotransmitter-based explanation, toward early changes in emotional and social processing and toward neural plasticity (Harmer et al., Lancet Psychiatry, 2017).
The human evidence for SSRIs
The largest synthesis in depression pooled 522 double-blind randomised trials covering 116,477 participants and 21 antidepressants, SSRIs among them. All 21 were more efficacious than placebo. In head-to-head trials, escitalopram and paroxetine were among the more effective drugs, and citalopram, escitalopram, fluoxetine and sertraline among the better tolerated (Cipriani et al., Lancet, 2018).
In generalised anxiety disorder, a network meta-analysis of 89 trials and 25,441 patients ranked escitalopram among the drugs more efficacious than placebo with relatively good acceptability. Sertraline and fluoxetine were also efficacious and well tolerated, though with smaller samples, and paroxetine was effective but less well tolerated (Slee et al., Lancet, 2019). The class carries a boxed warning on suicidality in children and young adults aged 18 to 24 (Edinoff et al., 2021).
What is Selank?
Selank is the synthetic heptapeptide Thr-Lys-Pro-Arg-Pro-Gly-Pro, developed from tuftsin, a peptide the body makes (Kolik et al., Bull Exp Biol Med, 2019). Its anti-anxiety mechanism research centres on two systems. It inhibited the enzymes that break down enkephalins in human plasma, and enkephalin half-life was shortened in patients with generalised anxiety (Zozulya et al., Bull Exp Biol Med, 2001). In radioligand work it acted as a positive allosteric modulator of GABA binding (Vyunova et al., Protein Pept Lett, 2018). It is introduced in more depth in Selank peptide: what the research covers.
Selank and serotonin
Selank’s effect on serotonin looks nothing like an SSRI’s. In a 2008 mouse study, it raised noradrenaline in the hypothalamus of two strains and, in the more anxious BALB/c strain, lowered serotonin and its metabolite 5-HIAA in the hippocampus, leaving them unchanged in the other strain; the authors read this as a sign of selective anxiolytic action (Narkevich et al., Eksp Klin Farmakol, 2008). In rats, Selank changed the expression of 45 neurotransmission genes within an hour, a panel that included serotonin and dopamine receptors (Volkova et al., Front Pharmacol, 2016). Rat work has also linked it to BDNF, a growth factor tied to neural plasticity, in the hippocampus and prefrontal cortex (Kolik et al., 2019).
The human studies of Selank
Selank’s clinical studies are in anxiety disorders, with benzodiazepines as comparators. In 2008, 62 patients with generalised anxiety disorder or neurasthenia received Selank (30) or medazepam (32); the anti-anxiety effects were similar, Selank added antiasthenic and psychostimulant effects, and serum enkephalin half-life rose during the Selank course (Zozulia et al., Zh Nevrol Psikhiatr Im S S Korsakova, 2008). In 2014, a study of 60 patients with anxiety-phobic and somatoform disorders reported a pronounced anti-anxiety effect lasting a week after the last administration, a mild nootropic effect and better quality of life (Medvedev et al., 2014). In 2015, adding Selank to phenazepam brought improvement sooner and reduced the benzodiazepine’s side effects (Medvedev et al., 2015). The full record is in Selank and anxiety: the research.

Where the two research literatures meet and part
- Condition studied: both have anxiety research, but SSRIs carry the depression evidence. We found no published controlled trial of Selank in major depressive disorder.
- Comparators: Selank was compared with benzodiazepines, not with SSRIs. We found no head-to-head trial between Selank and any SSRI.
- Time course: SSRI research deals with a delayed clinical onset; Selank’s 2014 study tracked an effect that persisted for a week after the course ended, a different kind of timing question.
- Systems: SSRI research is anchored in serotonin; Selank’s in enkephalins and GABA, with serotonin changes reported only in animal work.

How is Selank supplied?
We supply Selank as a 10 mg lyophilised vial, as the ready-mixed Selank Nasal Spray (a metered pump delivering 0.175 mL per press) and in the Sleep & Calm Spray Stack with DSIP. Vials are reconstituted with bacteriostatic water; the Selank reconstitution guide has a calculator, and How long does Selank last? covers storage. Certificates of analysis are on the lab reports page, and every order ships from within Canada by Canada Post Xpresspost.
Selank is supplied for laboratory research use only. The studies above were clinical research using pharmaceutical preparations. SSRIs are prescription medicines, and nothing here compares them with Selank for personal use.
Frequently asked questions
What is the difference between Selank and an SSRI?
SSRIs are approved antidepressants that block the serotonin transporter and raise synaptic serotonin. Selank is a seven-amino-acid tuftsin-based peptide whose research points to enkephalin-degrading enzymes and allosteric modulation of GABA binding rather than serotonin reuptake.
Does Selank affect serotonin?
In a 2008 mouse study, Selank lowered serotonin and its metabolite in the hippocampus of the more anxious BALB/c strain and did not change them in another strain. That is a different pattern from an SSRI, which raises synaptic serotonin.
Has Selank been compared with SSRIs?
We found no published head-to-head trial. Selank’s clinical comparators were the benzodiazepines medazepam and phenazepam, and its studies were in anxiety disorders rather than major depression.
Has Selank been studied for depression?
Its human studies were in generalised anxiety disorder, neurasthenia and anxiety-phobic and somatoform disorders. We found no published controlled trial of Selank in major depressive disorder.
