- In 49 human skin samples from newborns to age 77, NAD+ levels fell with age in both males and females (Massudi, PLoS One, 2012).
- In healthy people aged 20 to 87, plasma NAD+ and NADP+ declined significantly with age (Clement, Rejuvenation Res, 2019).

What is NAD+?
NAD+ (nicotinamide adenine dinucleotide) is the coenzyme at the centre of cellular energy production. It carries electrons in mitochondrial respiration and oxidative phosphorylation, and it is the sole substrate for the DNA repair enzyme PARP and for the sirtuin family of NAD-dependent deacetylases (Massudi, 2012). Research areas include cellular energy, mitochondrial function, DNA repair, sirtuin activity, oxidative stress, mental clarity and healthy aging.
Why is NAD+ studied as a nasal spray?
The nasal cavity gives direct access to the olfactory epithelium, and the 2026 study made that its target. Intranasal NAD restored olfactory function in an anosmia model, and in cultured human olfactory stem cells NAD raised markers of olfactory sensory neurons and upregulated neuronal differentiation genes (Yoo, 2026). What does the research on NAD+ and aging show?
Human tissue and blood studies both track NAD+ downward with age. In the skin-sample study, PARP activity rose with age in males and correlated inversely with tissue NAD+, and the authors proposed that NAD+ depletion may limit energy production, DNA repair and genomic signalling (Massudi, 2012). In plasma, NAD+, NADP+ and NAAD all declined with age, while NADH, NADPH and nicotinamide rose (Clement, 2019).
That decline is why NAD+ and its precursors are studied for raising NAD+ availability. A randomised, double-blind, placebo-controlled crossover trial found that chronic nicotinamide riboside, an NAD+ precursor, effectively stimulated NAD+ metabolism in healthy middle-aged and older adults (Martens, Nat Commun, 2018). A 2024 systematic review of 10 randomised trials with 489 participants, covering chronic fatigue syndrome, older adults, Parkinson’s disease, overweight, postmenopausal prediabetes and Alzheimer’s disease, found NADH and its precursors well tolerated (Gindri, Am J Physiol Endocrinol Metab, 2024).
Is NAD+ the same as NADH or nicotinamide riboside?
No, though all three belong to the same system. NAD+ is the oxidised coenzyme itself, the form our spray contains. NADH is its reduced form, the one used in the chronic fatigue study, where no severe adverse effects were observed (Forsyth, 1999). Nicotinamide riboside is a vitamin precursor that the body converts into NAD+ (Martens, 2018). Studies on precursors measure how much NAD+ the body makes from them; our spray supplies NAD+ itself rather than a precursor.
