- In 24 healthy volunteers, intranasal Semax changed the brain’s default mode network on fMRI compared with placebo, with scans at 5 and 20 minutes (Lebedeva, Bull Exp Biol Med, 2018).

Why are nootropic peptides studied as nasal sprays?
The nasal route is how much of the Semax research was done. In a resting-state fMRI study, healthy volunteers received intranasal Semax or placebo, and the Semax group showed a larger frontal (medial frontal cortex) component of the default mode network within 20 minutes (Lebedeva, 2018). In an animal model, intranasal Semax raised BDNF protein in the basal forebrain within 3 hours (Dolotov, J Neurochem, 2006).

What is Semax nasal spray studied for?
Semax is a seven-amino-acid analogue of the 4 to 10 fragment of adrenocorticotropic hormone (ACTH), studied for attention, memory, learning and neuroprotection. Its central research thread is brain-derived neurotrophic factor (BDNF). In 110 patients after ischaemic stroke, Semax raised plasma BDNF, which stayed high for the whole study, and improved final Barthel Index scores, a standard measure of independence in daily activities (Gusev, 2018).

What is Selank nasal spray studied for?
Selank is a seven-amino-acid analogue of tuftsin, studied for anxiety, the stress response and a calmer state without sedation. In 62 patients with generalised anxiety disorder and neurasthenia, Selank matched the anxiolytic effect of medazepam and also had antiasthenic and psychostimulant effects (Zozulia, Zh Nevrol Psikhiatr, 2008). In 60 patients compared against phenazepam, its anxiolytic effect lasted a week after the last administration (Medvedev, 2014).

In a double-blind, placebo-controlled study of 14 middle-aged people with chronic insomnia, DSIP substantially improved night sleep, the effect held into the first placebo night afterwards, and daytime alertness and performance rose significantly (Schneider-Helmert, 1987).
