Findings
- In 18 premenopausal women with sexual arousal disorder, a single intranasal administration of PT-141 meant more women reported moderate or high sexual desire than after placebo (Diamond, J Sex Med, 2006).
- In healthy men and men with erectile dysfunction, intranasal PT-141 produced a statistically significant erectile response compared with placebo, with the first erection at about 30 minutes (Diamond, Int J Impot Res, 2004).
- Intranasal kisspeptin rapidly raised luteinising hormone in healthy men and women under randomised, double-blind, placebo-controlled conditions, with no side effects or adverse events encountered (Mills, EBioMedicine, 2025).
- In 29 healthy couples, intranasal oxytocin increased the intensity of orgasm and contentment after intercourse (Behnia, Horm Behav, 2014).

Why are libido peptides studied as nasal sprays?
For all three compounds, the nasal route sits close to the centre of the human research. The early human PT-141 trials used an intranasal form in both men and women (Diamond, 2004 and 2006). Most published human oxytocin research delivers it as a nasal spray, including the study that found intranasal oxytocin caused a substantial increase in trust between people (Kosfeld, Nature, 2005).
Kisspeptin joined them more recently. In a 2025 study, kisspeptin-54 was delivered effectively by nasal spray, stayed stable for up to 60 days at 4 °C, and raised luteinising hormone in healthy volunteers and in patients with hypothalamic amenorrhoea (Mills, 2025). What is PT-141 nasal spray studied for?
PT-141 (bremelanotide) is a synthetic melanocortin analogue that acts at the MC3 and MC4 receptors. It works on desire pathways in the brain rather than on blood flow, which is why it is studied in both men and women. In the 2006 study, women who attempted intercourse within 24 hours of PT-141 were significantly more often satisfied with their level of arousal than after placebo (Diamond, 2006).

What is oxytocin nasal spray studied for?
Oxytocin is the hormone studied for social bonding, trust, the stress response and relaxation. In the couples study, men reported higher sexual satiety after intercourse with oxytocin, and women felt more relaxed, with subgroups better able to share sexual desires and empathise with their partners (Behnia, 2014). What is Kisspeptin nasal spray studied for?
Kisspeptin is the natural hormone at the top of the reproductive axis: it prompts the release of LH and FSH, which drive sex-hormone production. Its research also reaches the brain circuits of desire. In 32 men with hypoactive sexual desire disorder, kisspeptin modulated brain activity across the sexual-processing network, raised penile tumescence by up to 56% more than placebo and increased happiness about sex (Mills, JAMA Netw Open, 2023). In 32 women with the same diagnosis, it modulated sexual and facial-attraction brain processing and was well tolerated (Thurston, JAMA Netw Open, 2022).
