- In 18 chronic insomniacs aged 29 to 83, a week of DSIP was followed by normal sleep patterns across the whole group by the end of the study (Schneider-Helmert, Eur Neurol, 1986).

Why use a ready-mixed spray for sleep research?

What is DSIP nasal spray studied for?
DSIP (delta sleep-inducing peptide) is a nine-amino-acid peptide first isolated in sleep research. It is studied for deep, delta-wave sleep, sleep architecture, circadian rhythm and stress hormones. In the 1986 sleep-laboratory study, the immediate effect of DSIP was largest in the older insomniacs, whose starting disturbance was twice as severe, and regression analysis tied the size of the effect to the degree of sleep disturbance (Schneider-Helmert, 1986).

What did the DSIP sleep studies measure?
The human DSIP work was done in sleep laboratories. The 1986 study recorded immediate effects at the end of one week of DSIP and late effects after a further week of follow-up, and split its 18 insomniacs into a middle-aged group (29 to 59) and an older group (60 to 83). In the middle-aged group, sleep reached normal values by the end of the DSIP week and stayed there through follow-up; in the older group, full normalisation came by the end of follow-up (Schneider-Helmert, 1986).
Circadian work adds the daytime side. With blood drawn every 30 minutes and simultaneous polygraphic recording, plasma DSIP peaked at 15:00 and was lowest at 01:00, and its rhythm closely followed body temperature (r2 = 0.66) (Friedman, 1994). That timing link is why circadian rhythm sits among the DSIP research areas.
Where does Selank fit in sleep and calm research?
Selank is a seven-amino-acid analogue of tuftsin, studied for anxiety, the stress response and a calmer state without sedation. That last point is why it pairs with DSIP: one compound for the sleep period, one for the calm, steady state around it. In the 2008 trial, Selank also raised the half-life of leu-enkephalin in patients with generalised anxiety disorder, a measure that had been low before treatment (Zozulia, 2008).
